3315 Confocal in vivo probing of proteins and water in the rabbit cornea

چکیده
Puroose: To develop an instrument with high spatial resolution probing chemical composition of comeal tissue in live animal model without interference of signals originating from adjacent tissues. Methods: A Raman probe for in viva studies was developed that has the required resolving power in space, obtained by a confocally placed fiber for transportation of the Raman signai. Resolving power in time. to suppres movement artifacts, was obtained by a microcope objective with a high light gathering power and a long working distance. Rzsults: fn viva Raman spectra of protein and water collected along the optical axis of the excitation beam were obtained from rabbit corneas at exposure times of 1 s at spatial intervals of 70 pm with a resolution in depth of 30 or 150 pm. Conclusions: The shallow probing depth of 30 pm proved to have enough spatial and temporal resolution to perform reliable measurements of the protein and water gradients in the comeal tissue in an animal model. However, for measurements where gradients are not relevant, the 150 pm probing depth, that requires less radiation energy, i:r adequate for intra comeal measurements.

Variations of the Normal Human Limbal Stem Cells Detected by In Vivo Confocal Microscopy
Background To report normal variations of the limbal structures using in vivo laser scanning confocal microscopy. Methods: This was a retrospective study of fourteen eyes from 11 healthy individuals. Confocal imaging of cornea and limbus was performed using Heidelberg Retina Tomograph III Rostock Corneal Module. Results: The typical structure of the palisades of Vogt (POV) was detected ...
متن کاملLoratadine- Loaded Thermoresponsive Hydogel : Characterization and Ex-vivo Rabbit Cornea Permeability Studies
Poor bioavailability of ophthalmic drops is mainly due to drainage through the nasal-lacrimal duct and a very low permeability through corneal epithelium. The aim of our study was to prepare and characterize an ocular hydrogel of loratadine, as an example of a lipophilic drug, to increase drug concentration and residence time at the site of action in the eye. In this study,a 23full factorial de...

متن کاملNoninvasive assessment of the hydration gradient across the cornea using confocal Raman spectroscopy.
PURPOSE The feasibility of Raman spectroscopy for the noninvasive assessment of axial corneal hydration was investigated. METHODS A scanning confocal Raman spectroscopy system, with an axial resolution of 50 microns, was used to assess noninvasively the water (OH-bond) to protein (CH-bond) ratio as a measure of the hydration in collagen-based phantom media and rabbit corneas. RESULTS Raman ...
